在不同温度处理下,测定了心叶赠(Nicotiama glutinosa)接种烟草花叶病毒(Tobacco Mosaic Virus,TMV)后生成的枯斑数.结果表明:随着温度升高,半叶平均枯斑数呈上升趋势.二者呈极显著相关(r=0.989,P=0.001〈0.01),且半叶平均枯斑数与处理温度之间存在显著的线性回归关系.  相似文献

将BnTR1基因转化到了本生烟中,发现与非转基因对照相比,转基因烟草对甘露醇和PEG模拟干旱表现出较强的抗性,尤其是PEG胁迫时,三个转基因株系在生物量的积累和整体长势方面都优于对照.干旱处理20d后复水并培养14d后,对照烟草全部枯死,但是所有转基因烟草都恢复生长直至开花结果.当把烟草叶片圆片浸泡在系列盐水中,盐浓度超过600mM时,对照烟草圆片受损白化,而转基因烟草圆片能保持较大面积的绿色。热激蛋白基因HSP90和HSP70在所有烟草中的表达量都有明显提高,其表达量的增加在对照和转基因株系中并无差异,但是在正常条件下不表达的HSF30和sHSP17.6基因在热激处理后都表达了,而且HSF30的表达量增加了50%,sHSP17.6的表达量增加了30%.  相似文献

Summary The study of malate dehydrogenase patterns in leaves ofNicotiana suaveolens, N. glutinosa and their interspecific hybrid has been carried out, in order to propose a model of its genetic control. Two possible explanations for the genetic control of malate dehydrogenase have been postulated forN. suaveolens, and at least two loci appear to be implicated in the control of this system inN. glutinosa.Acknowledgments. 漂盘培育云烟87和三生烟,苗期接种TMV后0h,12h,24h,36h,48h,60h和72h分别取样测定其叶片和根系中PAL的活性,并以未接种TMV烟苗叶片和根系中PAL的活性为对照.结果表明:烟草与TMV非亲和性互作时,叶片和根系中PAL活性上升速度比烟草与TMV亲和性互作时PAL活性上升速度快;烟草与TMV无论是亲和性互作还是非亲和性互作,烟草叶片中PAL活性升高速度快于根系中PAL活性上升速度.  相似文献

Leaves of transgenic sweetgum (Liquidambar styraciflua) trees that expressed tobacco anionic peroxidase were compared with leaves of L. styraciflua trees that did not express the tobacco enzyme. Leaves of the transgenic trees were generally more resistant to feeding by caterpillars and beetles than wild-type leaves. However, as for past studies with transgenic tobacco and tomato expressing the tobacco anionic peroxidase, the degree of relative resistance depended on the size of insect used and the maturity of the leaf. Decreased growth of gypsy moth larvae appeared mainly due to decreased consumption, and not changes in the nutritional quality of the foliage. Transgenic leaves were more susceptible to feeding by the corn earworm, Helicoverpa zea. Thus, it appears the tobacco anionic peroxidase can contribute to insect resistance, but its effects are more predictable when it is expressed in plant species more closely related to the original gene source. 分别构建了iaaM、iaaM和ipt共表达植物转化载体,农杆菌介导转化烟草,RT-PCR鉴定转基因株系,ELISA方法分析叶片IAA、IPA和ZR含量.结果表明:叶片IAA含量高低依次为株系iaaM10、iaaM_ipt3和野生型;而细胞分裂素含量高低依次为iaaM_ipt3、野生型和iaaM10.随着内源IAA水平升高,气孔密度减小、保卫细胞长度增加、栅栏组织细胞增大和叶片厚度增加.讨论了生长素和细胞分裂素含量对叶片细胞生长发育的影响.  相似文献

PCR扩增利用吲哚乙酰胺水解酶基因(iaaH)全长序列,构建iaaH基因序列正、反向连接的的双元载体,通过农杆菌LBA4404介导,转化携带pTiC58 T-DNA的烟草叶片,获得携带正、反向iaaH序列的转基因植株.ELISA分析生长素的结果表明,转基因植物叶片的内源生长素含量明显降低,仅为pTiC58 T-DNA转化植株的56%,说明转基因烟草中发生iaaH基因转录后沉默,为利用该基因序列启动RNA干扰(RNAi),抑制冠瘿瘤病发生提供了实验依据.  相似文献

FtsZ protein plays an important role in the division of chloroplasts. With the finding and functional analysis of higher plant FtsZ proteins, people have deepened the understanding in the molecular mechanism of chloroplast division. Multiple ftsZ genes are diversified into two families in higher plants, ftsZ1 and ftsZ2 . On the basis of the research on ftsZl family, we analyzed the function of NtFtsZ2-l gene in Nicotiana tabacum . Microscopic analysis of the sense and antisense NtFtsZ2-l transgenic tobacco plants revealed that the chloroplasts were abnormal in size and also in number when compared with wild-type tobacco chloroplasts. Our investigations confirmed that the NtFtsZ2-l gene is involved in plant chloroplast division.  相似文献
